Monday 9th March - Introduction to Flow Cytometry: The Foundation Block is aimed at those new to flow cytometry and those wanting to understand the basics of the technology before attending further Building Blocks. This can be taken as a standalone one day course or can be taken as a building block for the next courses. The course starts with an introduction to this quickly evolving technology, its importance and resources that are available. One of the main aims of this course is to give the delegate a firm understanding of fluorescence, fluorochromes and the workings of a cytometer. This is followed by talks on fluorescence compensation, sample preparation, basics of experimental design, data analysis and reporting your flow cytometry data.


Tuesday 10th March - Advanced Flow Cytometry: This interactive course builds on the Introductory course and goes into greater details on how a cytometer works, best practices for running cytometry experiments, experimental design, panel design, compensation and data spreading and presentation of data. As cytometers become capable of measuring an increasing number of fluorochromes, experimental design becomes more complicated. It requires knowledge of the fluorochrome, the biology (antigen density, co-expression) and the sample preparation as well as as a greater understanding about the cytometer. In this one-day course we will look at the factors that need to be taken into account to build multicolour panels. We will do several interactive panel designs where we will use this knowledge to design panels of 6-10 and discuss panels with larger numbers of fluorochromes.


Wednesday 11th & Thursday 12th - Practical Cell Sorting: Populations detected by analytical cytometry may be retrieved using droplet deflection cell sorting. Successful sorting requires knowledge of the sorting process and how to adjust parameters to maximise purity or yield There may be special considerations of downstream applications eg single cell sorting for RNAseq experiment. This two-day course will introduce delegates to the theory of cell sorting but will also include a practical element where we will using a range of cuvette-based system and a stream-in-air systems (including FACSAria Fusion, FACSJazz and Bigfoot sorters). There will also be exposure to the range of sorters avaliable at Babraham Institute during a lab tour including the new BD FACSDiscover S8 spectral sorter with imaging capabilities; and Cytek Aurora CS.


Friday 13th March - Practical Data Analysis: Data analysis of flow data files can be daunting. Where do I set regions and gates? Which metric do I use? In this one-day course we will guide delegates through the minefield of data analysis from simple gating to high dimensional analysis. This one-day course will introduce delegates to the analysis of flow data. We will cover the plot types, setting compensation, regions and gates, the metrics that can be derived, and data presentation. We will illustrate all these with data files that will be available to delegates.

PLEASE NOTE: YOU ARE REQUIRED TO BRING A LAPTOP TO THE DATA COURSE AND PLEASE DOWNLOAD THE LATEST VERSION OF FLOWJO ON TO IT. WILL WE SEND DETAILS OF A TEMPORARY FLOWJO LICENCE TO BE USED ON THE COURSE NEARER THE TIME OF THE COURSE. IF YOU DO NOT HAVE A LAPTOP PLEASE LET US KNOW IN ADVANCE AND WE WILL BE ABLE TO PROVIDE ONE.

The in person courses includes refreshments, including lunch, morning and afternoon tea and coffee and course notes sent in advance.
