Title: Seeing is believing? Exploring Ultrahigh-Sensitive Spatial Proteomes Enriched with Visual Context

Abstract: Advances in spatial proteomics have transformed our ability to explore the molecular architecture of tissues with ultrahigh sensitivity and unparalleled precision. Mapping the spatial distribution of proteins within cells and tissues is crucial for understanding their influence on cellular behaviour. Spatial proteomics, recognized as the 2024 "Method of the Year," leverages AI-driven approaches to integrate microscopy and mass spectrometry data enabling precise annotation of protein function and localization. The high-resolution visually supported proteome data offers deeper insights into biological processes operative in specific cellular subsets. In my presentation, I will provide the potential of combining ultrahigh-sensitivity spatial proteomics with visual context to uncover critical insights into health and disease. Using the persistence of SARS-CoV-2 spike protein at the skull-meninges-brain axis as a case study, we demonstrate how spatially resolved proteomics revealed the accumulation of spike protein in specific tissue compartments. By integrating these findings with 3D imaging, we contextualized protein distribution and its association with neurodegeneration. Additionally, I will discuss the future applications of these technologies in fields such as immunology and cancer research, where spatial resolution and proteomic sensitivity are paramount. By combining the power of “seeing” with the precision of molecular “believing,” this approach promises to unveil new dimensions of biology with profound implications for biomedical research. At Yale University, we utilize insights gathered from spatial proteomics to conduct immunopeptidome investigations at spatial resolution.
