Abstract:
NanoTemper Technologies has developed a novel approach to the characterization of molecular interactions: Spectral Shift. This new technology is the driving engine for our latest product releases Dianthus and Monolith X. Both instruments feature a combination of the new isothermal spectral shift detection with the established MST/TRIC technology. The instruments enable characterization of in solution interactions for a wide range of biomolecules, even for challenging samples such as PROTACs, membrane proteins, intrinsically disordered proteins, and cell lysates. The instruments enable mass-independent, in-solution measurements with lowest sample consumption and even in the presence of aggregation. Dianthus and Monolith X can not only be used to validate your results from other biophysical methods but also to characterize your most challenging interactions. NanoTemper Technologies has developed the Prometheus which the scientific community has chosen because it consistently delivers high-resolution data that reveals details in proteins and formulations that are missed by other technologies. The latest model (Prometheus Panta) is equipped with the current gold standard for protein stability characterization (label free nanoDSF + backreflection), and a novel type of DLS/SLS enabling new experimental possibilities.
Besides standard applications, the new type of DLS/SLS now allows dynamic light scattering (DLS) experiments in tandem with static light scattering (SLS), label free nano-differential scanning fluorimetry (nanoDSF) and backreflection, along an entire thermal ramp, to gain information on conformation, thermal stability, turbidity, radius, size distribution and more, on your protein of interest or formulation.
