The sudden loss of movement after a stroke or spinal cord injury (SCI) is life-changing. In a moment, the injured person can lose independence, becoming reliant on care-givers for daily activities. Paralysis after stroke and SCI is a major impetus to study motor system plasticity, to develop novel repair strategies that rewire the damaged motor system and promote functional recovery. The motor cortex and the corticospinal tract are key to producing voluntary movements. In health and after injury, the spinal cord is an important site for activity-dependent plasticity of the corticospinal tract. My laboratory uses animal models to study activity-dependent axon outgrowth and synaptic plasticity that can be augmented by motor cortex and spinal cord neuromodulation to promote motor function after injury. My presentation will focus on the corticospinal motor system. I will talk about our research on how motor cortex neuromodulation can be used to upregulate molecular programs for axon growth and synaptogenesis and, in turn, promote activity-dependent corticospinal axon sprouting, synaptogenesis, and long-term potentiation (LTP) at the corticospinal tract-spinal neuron synapse in the spinal cord. I will focus on several rodent injury models, and show how neuromodulation approaches can be used to repair the damaged corticospinal system, reestablish key physiological processes, and promote recovery of skilled movements after SCI.
